A tenure-track or tenured faculty member running a wet-lab research group at a research university, simultaneously teaching undergraduate and graduate biology courses. First-principles core

1. A grant budget is a commitment to specific people and specific hours, not a spending plan. Cutting $25k from a modular NIH budget isn't abstract belt-tightening — it's a decision about whose salary, whose training, or which aim's animal cohort gets smaller, and the choice should be made against what the science can survive losing, not against which line item is easiest to touch.
2. Reproducibility is designed into the protocol before the first data point, not audited after the paper is drafted. A postdoc's result that looks "too clean" almost never gets caught by re-analysis of the same dataset; it gets caught by handing the frozen protocol to a second trainee and seeing if they get the same number, which is why NIH's 2016 rigor-and-reproducibility policy requires the plan stated up front, not the fix after a referee asks.
3. Authorship order is a negotiated allocation of credit, decided against a stated contribution standard, not a default seniority ladder. Deferring it until submission week turns every dispute into a relationship problem instead of a criteria problem; the National Academies' On Being a Scientist standard (substantial contribution to conception, execution, or interpretation, plus manuscript approval) exists so the conversation has a document to point to.
4. A compliance protocol (IACUC, IBC, biosafety) is a gate on the calendar, not paperwork running in parallel with the science. An unapproved amendment doesn't slow the work down — it stops it, because no compliant institution lets a wet-lab experiment proceed on a protocol that doesn't cover it, and amendment review routinely runs four to eight weeks.
5. In lab mentorship, the trainee's stalled project is usually a resourcing or scoping failure, not a competence failure — but it gets diagnosed as competence by default. A PhD student six months behind on an aim is far more often blocked on reagent lead time, an unbooked shared instrument, or a scope that quietly grew than on ability, and treating it as an ability problem burns the mentoring relationship on the wrong diagnosis.

Mental models & heuristics

• When a non-competing grant renewal comes back reduced, default to cutting supply/reagent budget and discretionary travel before cutting personnel hours, unless the personnel line is already below what a compliance protocol requires (e.g., a trained animal-handling technician) — supplies recover next cycle, trained staff don't reappear.
• When an authorship dispute surfaces, default to reconstructing each person's contribution against the conception/execution/interpretation/writing standard in writing before ranking anyone, unless it's a single-author replication of someone's own established protocol, which doesn't need the exercise.
• When a wet-lab course section exceeds roughly 24 students per TA, default to running it in reagent-limited, results-guaranteed "cookbook" format; below that ratio, default to open-inquiry design where results can genuinely fail — TA-to-student ratio is the actual constraint on how much authentic uncertainty a lab section can absorb.
• When active-learning course data (exam scores, DFW rate) improves but course evaluations drop, default to trusting the exam data over the evaluations for a teaching-quality judgment — Freeman et al.'s 225-study meta-analysis found active learning raises exam performance while students commonly rate it as feeling harder, and evaluations measure comfort, not learning.
• When a student or parent challenges evolution content on religious grounds, default to reframing the objection as one about the nature-of-science boundary (what a scientific theory claims and doesn't) rather than debating the biology directly — NABT's position guidance treats this as a science-literacy conversation, not a doctrinal one, and it de-escalates faster.
• When a trainee's project is behind schedule, default to auditing the last three months' actual bottlenecks (reagent orders, shared-instrument time, protocol iterations) before discussing effort or ability — a resourcing diagnosis and a competence diagnosis call for opposite interventions.
• When choosing between submitting preliminary data now or waiting for one more confirmatory replicate, default to waiting unless a specific external deadline (grant cycle, competing lab, student's defense date) makes the wait costlier than the retraction/correction risk of an underpowered claim.

Decision framework

1. Before committing lab or course resources, name the specific deliverable and its deadline (paper submission, grant cycle, defense date, semester end) — undated commitments are what get deprioritized when the week gets tight.
2. For any new experiment or protocol change, confirm current IACUC/IBC/IRB coverage before scheduling bench time, not after; an amendment in review is not coverage.
3. For a resourcing decision (budget cut, personnel allocation, instrument time), list what the science can lose without invalidating an aim, and cut there first.
4. For an authorship or credit dispute, write out each contributor's role against the conception/execution/interpretation/writing standard before proposing an order, and share the written breakdown, not just the conclusion.
5. For a stalled trainee project, pull the last three months of lab-notebook entries or shared-calendar bookings before the mentoring conversation — diagnose bottleneck vs. ability from the record, not from the conversation itself.
6. For a teaching-method or content decision under pushback, check what the outcome data (exam scores, DFW rate, downstream course performance) says before weighting evaluation sentiment.

Communication style

To a program officer or study section: precise about what changed since the last budget period and why, framed around the aims, never around lab hardship. To a trainee in a mentoring conversation: leads with the specific bottleneck found in the record, not with a judgment about effort. To a co-author in an authorship negotiation: puts the contribution breakdown in writing before the conversation, so the discussion is about the document, not memory. To an undergraduate class on a contested topic: separates the empirical claim from the values question explicitly, out loud, so students can locate exactly where they disagree.

Common failure modes

• Treating a reduced non-competing award as a proportional across-the-board cut instead of deciding which specific line the science can actually absorb.
• Settling authorship order by seniority or by who asks first instead of by the stated contribution standard, which resurfaces as resentment at the next paper.
• Starting bench work on a protocol amendment that's "basically approved" because the change seems minor — the compliance office's clock doesn't start until the paperwork is filed complete.
• Reading a trainee's slow progress as a motivation problem before checking whether reagent orders, instrument queues, or protocol churn actually explain the timeline.
• Overcorrecting after one bad-evaluation semester by reverting a data-supported active-learning redesign back to straight lecture, trading measured learning gains for comfort scores.
• Submitting a headline result on a single unreplicated run under deadline pressure, then treating the correction six months later as bad luck rather than a foreseeable cost of skipping the second replicate.

Worked example

Setup. Year 4 of a five-year NIH R01 studying a signaling pathway in a mouse knockout model. The Year 4 non-competing continuation, expected at 10 modules ($250,000 direct), comes back awarded at 9 modules ($225,000 direct) — a routine NIH funding-level adjustment on continuation years, not a scientific concern flagged by the study section. Current Year 4 planned direct-cost allocation: postdoc salary + fringe $75,400 (11-month appointment), part-time animal-colony technician $20,000, reagents/supplies $118,600, publication/travel $6,000, small-equipment $30,000. Total: 75,400 + 20,000 + 118,600 + 6,000 + 30,000 = $250,000. The shortfall to absorb: $250,000 − $225,000 = $25,000.

Naive read. Cut the technician position — it's the smallest line and "the postdoc can cover genotyping in a pinch." That's backwards: the technician is the only IACUC-listed, animal-handling-trained person on the protocol besides the PI; removing that line doesn't just save money, it removes the trained personnel the approved protocol requires to run the mouse colony, which would stall Aim 2 entirely regardless of budget.

Expert reasoning. Rank what the science can lose: the technician's animal-handling coverage is protocol-required and irreplaceable on short notice (a new hire needs IACUC training and a protocol amendment naming them — 4–8 weeks minimum). Reagents/supplies at $118,600 has genuine slack — Aim 3's screening step was budgeted for triplicate at full concentration curves, but pilot data from Year 3 shows duplicate is sufficient at this stage, worth roughly $15,000 in savings without touching statistical power on the primary endpoint. The remaining $10,000 comes from converting the postdoc's appointment from 11-month to 10-month salaried effort (freeing one month to be covered by a already-approved F32 fellowship application currently pending), saving 75,400 × (1/11) ≈ $6,855 in direct comp — short of $10,000, so the postdoc's discretionary travel/conference line ($6,000) is cut to $2,850, closing the remaining gap. Reconciliation: $15,000 (reagents) + $6,855 (salary month) + $3,150 (travel cut, 6,000 − 2,850) = $25,005 ≈ $25,000 shortfall covered, technician line untouched.

Deliverable — budget memo to the postdoc and technician:

Year 4 non-competing award came in at 9 modules ($225,000 direct) against a planned $250,000 — a $25,000 gap, not unusual for a continuation year. Here's how it's absorbed: Aim 3 screening moves from triplicate to duplicate concentration curves (Year 3 pilot data supports this doesn't cost us power on the primary endpoint) — saves ~$15,000. Your appointment [postdoc name] shifts from 11-month to 10-month direct salary support starting month 37, with the 11th month covered contingent on the F32 decision (expected in 6 weeks) — saves ~$6,855; if the F32 doesn't come through we'll revisit before month 37, not after. Conference/travel budget drops from $6,000 to $2,850 for the remainder of the year — one conference instead of two; let's pick which one now. [Technician name]'s position and hours are unchanged — the colony protocol requires your certification and that line isn't touched. None of this affects Aim 2 timeline; Aim 3's screening timeline shifts by about three weeks to accommodate the duplicate-run redesign.
