Overview

protocol_video_matching bridges the physical bench and the digital protocol by continuously aligning a first-person XR headset video stream (e.g., Meta Quest, HoloLens 2, Magic Leap) against structured protocol text in real time. The skill parses each protocol step into a semantic action graph, tracks operator gestures and reagent interactions through a Vision-Language Model (VLM), detects when execution diverges from the ground-truth procedure, and surfaces instant corrective guidance as spatial overlays — turning every researcher into a compliant, self-auditing one-person lab.

When to Use This Skill

Use this skill when any of the following conditions are present:

• Live XR-assisted experiments: An operator wearing a first-person XR headset is executing a wet-lab protocol (PCR, cell culture, CRISPR editing, RNA extraction, Western blot, etc.) and needs real-time step-by-step guidance or compliance validation.
• Protocol compliance auditing: A lab manager needs post-hoc or live documentation showing whether a protocol was followed exactly — including timing, reagent volumes, temperature set-points, and action sequence.

• Protocol versioning: Tracks diff between protocol revision A and B and highlights changed steps for the operator

2. Real-Time Video Frame Analysis via VLM

Processes the live XR video stream to extract semantic lab actions at up to 10 fps:

• Action recognition: Detects pipetting, centrifuge loading, tube labeling, reagent addition, gel loading, microscope focusing, and 40+ common wet-lab gestures using a fine-tuned VLM (e.g., GPT-4o Vision, Gemini 1.5 Pro, LLaVA-Med)

• Deviation taxonomy: Classifies each divergence as one of — STEP_SKIPPED, STEP_OUT_OF_ORDER, WRONG_PARAMETER (volume, time, temperature, reagent), TIMING_VIOLATION, LABELING_ERROR, EQUIPMENT_MISMATCH
• Severity scoring: Assigns a severity level (CRITICAL, MAJOR, MINOR) based on downstream impact on experiment validity
• Compliance score: Emits a rolling 0–100 compliance score per protocol section and a final session score suitable for ELN attachment
• Near-miss detection: Flags actions that are almost correct but fall outside tolerance bounds (e.g., 48 µL dispensed when 50 µL required, within 5% → MINOR WARNING)

4. Real-Time XR Corrective Guidance Overlay

Delivers corrections and confirmations as spatial intelligence anchored to the operator's field of view:

• Step completion check-off: A spatial checklist hovers at the bench edge; confirmed steps are ticked off automatically upon VLM validation
